  • Step One: Resin Swelling & Conditioning 

    Purpose: Enable resin accessibility and mass transfer for consistent coupling

    Typical Solvents: DCMTHFDMF, NMP, DMAc, EA. Custom blends are available upon request. 

  • Step Two: Deprotection 

    Purpose: Remove the temporary Fmoc protecting group to expose the reactive amine 

    Typical Blends: 20%(v/v) piperidine solution in solvent of choice (DCMTHFDMF, NMP)

  • Step Three: Coupling (repeat cycle) 

    Purpose: Attach the next amino acid with high efficiency and minimal side reactions

    Typical Solvents: Polar aprotic solvents (often DMF/ NMP/ DMAc) for consistent solubility and kinetics

  • Step Four: Wash Cycles 

    Purpose: Clear residual reagents/byproducts between steps to protect sequence fidelity

    Typical Solvents: DMF/NMP/DMAc/DCM/EA 

  • Step Five: Cleavage & global deprotection 

    Purpose: Release peptide from resin and remove side-chain protections

    Reagent: TFA 

  • Step Six: Precipitation / Isolation 

    Purpose: Crash out peptide and remove cleavage components

    Typical solvents: Diethyl Ether, Isopropyl Ether, MTBE  

  • Step Seven: Purification & Analytics 

    Purpose: Isolate target purity and support QC testing
